青岛地区3株犬细小病毒全基因组分析

摘要(Abstract):

为了探明青岛地区免疫过犬细小病毒(CPV)疫苗的犬因犬细小病毒感染死亡的原因,本研究采用全基因序列测定、系统进化树构建、基因分型和氨基酸分析的方法,对从临床发病犬中分离的3株CPV进行分析。结果显示,本实验室于2012年和2015年分离到的2株病毒(CPV-QD12和CPV-QD15)为new CPV-2a型,本研究分离到的1株病毒(CPV-QD20)为CPV-2c型;3株CPV在VP2的第5、267、297、324、370、426、440位氨基酸,NS1的第60、544、545、572、624、630位氨基酸发生了非同义替代。目前广泛使用的进口CPV弱毒活疫苗的基因型为原始的CPV-2型,与CPV流行毒株的基因型存在差异,这可能是造成CPV临床免疫失败的重要原因。因此,为了提升CPV疫苗的保护效率,建议使用CPV流行毒株研发CPV疫苗。
To investigate the cause of death in dogs immunized with canine parvovirus(CPV) vaccine in Qingdao, this study carried out the whole gene sequencing, evolutionary tree construction, genotype and amino acid analysis of 3 CPV epidemic strains isolated from clinically diseased dogs in Qingdao area. The results showed that the two strains(CPV-QD12 and CPV-QD15) previously isolated in our laboratory in 2012 and 2015 were new CPV-2a type and one virus strain(CPV-QD20) isolated in 2020 was CPV-2c type. The three CPV strains had non-synonymous substitutions at amino acids 5,267,297,324,370,426,and 440 of VP2 and amino acids 60,544,545,572,624,and 630 of NS1. The genotype of the currently widely used imported CPV attenuated live vaccine is the original CPV-2 type, which differs from the genotypes of the CPV epidemic strains, and may help explain for immune failure. Therefore, in order to improve the protective efficiency of CPV vaccine, it is recommended to use vaccine developed from CPV epidemic strains.
