中国大鲵出血症发病原因及致病菌基因组分析

摘要(Abstract):

为了探明中国大鲵出血症发病原因,本试验采集发病中国大鲵样本,运用16S rDNA序列分析结合细菌生化鉴定,对致病菌进行初步鉴定,利用Illumia测序方法,对致病菌进行全基因组测序,并对基因功能进行注释和预测。结果显示:该病发病原因为细菌感染,致病菌为摩氏摩根菌,对卡那霉素最敏感;该病原菌的基因组测序结果显示,其基因组长度为3.82 Mb,平均G+C含量为51.05%,含有3 732个基因,平均基因长度为895 kb;含有82个tRNA,总长度为6 439 bp,占基因总长度的0.16%,含有22个rRNA,总长度为31 986 bp,占基因总长度的0.83%;共计3 664个蛋白基因获得NR功能注释,并且注释得到1 780条GO功能条目和2 328条KEGG通路条目。本试验结果为中国大鲵出血症的防控和致病机制的研究提供了数据支持。
In order to investigate the pathogenesis of hemorrhagic disease in Andrias davidianus, this study collected samples from diseased Andrias davidianus, and preliminarily identified the pathogenic bacteria by using 16S rDNA sequencing in combination with biochemical tests. The Illumina platform was employed to sequence the complete genomes of the pathogenic bacteria and several bioinformatic tools were used to annotate and predict the gene functions. The results indicated that the cause of this disease was bacterial infection, and the pathogenic bacteria were identified as Morganella morganii, and were kanamycin sensitive. The complete genome harbored a 3.82 Mb chromosome with G+C content of 51.05%, and included 3 732 genes(average gene length of 895 kb), 82 tRNAs(total length of 6 439 bp) and 22 rRNAs(total length of 31 986 bp). Among the 3 664 protein-coding genes, 1 780 GO entries and 2 328 KEGG pathways were annotated. This study provides useful data for further analysis of the pathogenesis and control of hemorrhagic disease in Andrias davidianus.
