牛源化脓隐秘杆菌的分离鉴定与黔中金荞麦体外抑菌试验

摘要(Abstract):

为分离致病性牛源化脓隐秘杆菌并探究黔中金荞麦对该菌的体外抑制作用,本试验对采集于贵州省凯里市某养殖场疑似患牛呼吸道综合征的病死牛肺组织进行细菌分离、形态学观察、生理生化鉴定、分子生物学鉴定、药敏试验和致病性试验,并运用打孔法探究黔中金荞麦提取物对分离菌株的体外抑制效果。结果显示,分离菌株为革兰阳性短棒状杆菌,结合生理生化和PCR鉴定结果,确定该菌为化脓隐秘杆菌,系统进化树分析也显示,分离菌株与化脓隐秘杆菌的模式菌株处于同一分支;该分离菌株可致10只小鼠于24 h内全部死亡;其对头孢他啶、利福平、青霉素等7种抗菌药敏感,对环丙沙星中介,对克林霉素、左氧氟沙星、庆大霉素等8种抗菌药耐药;黔中金荞麦不同组织部位、不同处理方法的提取物对该分离菌株均具有一定抑制效果,其中地下部分70%乙醇超声30 min提取物的抑菌效果最好,抑菌圈直径为(28.13±0.25) mm,与头孢喹肟的抑菌效果相当。本试验首次从贵州地区分离到致病性牛源化脓隐秘杆菌,黔中金荞麦对该菌具有良好的体外抑制效果。本试验结果可为该肉牛养殖场疫病防控以及牛化脓隐秘杆菌感染的治疗和预防提供参考依据。
In order to isolate the pathogenic Trueperella pyogenes from bovine and investigate the in vitro inhibitory effect of Fagopyrum dibotrys Qian Zhong on T.pyogenes,this study conducted bacterial isolation,morphological observation,physiological and biochemical identification,molecular identification,drug sensitivity test and pathogenicity test of lung tissues collected from dead cattle suspected of respiratory syndrome in a breeding farm in Kaili City,Guizhou Province,and utilized punching method to assess the in vitro inhibitory effect of Fagopyrum dibotrys Qian Zhong extracts on the isolated strain.The results showed that the isolated strain was a Gram-positive short bacillus,and combined with the physiological and biochemical and PCR identification results,the bacterium was identified as Trueperella pyogenes.Phylogenetic tree analysis showed that the isolated strain was in the same branch as the model strain of T.pyogenes.The isolated strain killed all 10 mice within 24 h,and sensitive to 7 kinds of antibacterial agents,such as ceftazidime,rifampicin and penicillin,intermediate by ciprofloxacin,and resistant to 8 kinds of antibacterial agents,such as clindamycin,levofloxacin and gentamycin.The extracts from different tissue parts and different treatment methods of Fagopyrum dibotrys Qian Zhong had certain inhibitory effects on the isolated strain,with the 70% ethanol ultrasonic 30 min extract from the underground part displaying the best inhibitory effect,and the diameter of the inhibitory zone of(28.13±0.25) mm,indicating a comparable antibacterial effect to cefquinome.Thus,this study reported the isolation of a pathogenic T.pyogenes strain from bovine in Guizhou Province for the first time,and demonstrated the inhibitory effect of F.dibotrys Qian Zhong on the isolated T.pyogenes strain.These data provide scientific support for the prevention and control of T.pyogenes infection in cattle farms.
