复方蒲蠊提取物对溃疡性结肠炎大鼠菌群的干预作用
Effects of Compound Dandelion and Periplaneta americana Extracts on Intestinal Microflora of Rats with Ulcerative Colitis

摘要(Abstract):

为了研究复方蒲蠊提取物(PL1、PL2)对溃疡性结肠炎(UC)大鼠肠道菌群的影响,本试验采用2,4-二硝基氯苯(DNCB)联合醋酸建立UC模型,正常对照组和模型组分别灌肠给予生理盐水,阳性对照组灌肠给予康复新液,PL1低、中、高剂量组和PL2低、中、高剂量组分别灌肠给予不同浓度的PL1和PL2。采用疾病活动指数(DAI)、病理组织学评分(HS)和结肠黏膜损伤指数(CMDI)来评估康复新液、PL1和PL2对UC的治疗作用;收集结肠粪便,采用实时荧光定量PCR(qPCR)法检测各组大鼠粪便样本中主要菌种数量的变化。结果显示,与模型组相比,给予康复新液、PL1和PL2能够降低大鼠的DAI(P<0.01)、CMDI(P>0.05)、结肠指数(P<0.05或P<0.01)和HS总评分(P<0.05或P<0.01),显著增加结肠长度(P<0.05或P<0.01)。实时荧光定量PCR结果显示,与正常对照组相比,模型组双歧杆菌和乳杆菌极显著减少(P<0.01),大肠埃希菌、肠球菌、拟杆菌和硫酸盐还原菌极显著增多(P<0.01);与模型组相比,给予康复新液、PL1和PL2后大鼠粪便样品中双歧杆菌和乳杆菌极显著增多(P<0.01),大肠埃希菌、拟杆菌、肠球菌和硫酸盐还原菌显著减少(P<0.05或P<0.01)。结果表明,复方蒲蠊提取物对UC大鼠有一定的治疗作用,可能与调节UC大鼠肠道菌群的组成,维持有益菌和致病菌的平衡有关。
To investigate the effect of compound Dandelion and Periplaneta americana extracts(PL1 and PL2) on ulcerative colitis(UC) rats and analyze its effects on its intestinal flora,this study established UC model by using 2,4-dinitrochlorobenzene(DNCB) combined with acetic acid.Saline was administered by clyster to the normal control group and the model group.In the positive control group,Kangfuxin liquid was given by clyster,the administration groups were given PL1 low-dose,PL1 medium-dose,PL1 high-dose and PL2 low-dose,PL2 medium-dose,PL2 high-dose by clyster.The therapeutic effects of Kangfuxin liquid,PL1 and PL2 on UC were evaluated by disease activity index(DAI),histopathological score(HS) and colon mucosa damage index(CMDI).Rectal feces were gathered and the changes of bacteria in fecal samples of rats were detected by quantitative real-time PCR(qPCR).The results showed that Kangfuxin liquid,PL1 and PL2 reduced the DAI(P <0.01),CMDI(P>0.05),colonic index(P<0.05or P<0.01) and HS total score(P<0.05 or P<0.01),and increased colon length(P <0.05 or P<0.01) compared with the model group.qPCR results showed that compared with the normal control group,Bifidobacterium and Lactobacillus were reduced(P<0.01),and Escherichia coli,Enterococcusi,Bacteroides and sulfate-reducing bacteria were increased(P<0.01) in the model group.After administration,the growth of Bifidobacterium and Lactobacillus in the rat fecal samples were significant increased(P<0.01),and the growth of E.coli,Bacteroides,Etterococcus and sulfate-reducing bacteria was markedly inhibited(P <0.05 or P <0.01) in Kangfuxin liquid,PL1 and PL2 groups compared with those in the model group.Thus,the compound Dandelion and Periplaneta americana extracts had a certain therapeutic effect on UC rats,which might be related to regulating the composition of intestinal flora of UC rats and maintaining the balance of beneficial and pathogenic bacteria.
