高原兔肠道菌群多样性分析Analysis of Gut Microbial Diversity in Lepus oiostolus

摘要(Abstract):
为探究高原兔(Lepus oiostolus)肠道菌群多样性,本试验利用16S rDNA高通量测序方法分析高原兔小肠和大肠部位菌群种类及相对丰度,同时利用不同培养基对高原兔新鲜粪便中的微生物进行分离培养,并进一步对获得的菌株进行耐药性分析。16S rDNA高通量测序结果显示,高原兔小肠和大肠部位菌群在菌门水平主要由拟杆菌门(Bacteroidetes)、厚壁菌门(Firmicutes)、放线菌门(Actinobacteria)、互养菌门(Synergistetes)、蓝细菌门(Cyanobacteria)组成。优势菌属主要包括肠杆菌属(Enterobacter)、理研菌科RC9肠道群(Rikenellaceae RC9 gut group)、拟杆菌属(Bacteroides)、克里斯滕森菌科R-7群(Christensenellaceae R-7 group)。基于细菌培养分离方法共分离到25株细菌,隶属于6属、9种。耐药性分析发现,分离菌株对3~27种抗菌药物表现出耐药性。其中2种潜在有益菌株对5种常见肠道致病菌表现出一定的抑制能力。本试验结果为深入了解高原兔肠道微生物组提供了基础数据。
To explore the diversity of gut microbiota of Tibetan hare(Lepus oiostolus), this study applied 16S rDNA high-throughput sequencing method to determine the species and relative abundance of microbiota in its small and large intestines, and utilized different culture media to isolate microorganisms from its fresh feces followed by drug resistance testing on the obtained strains. The results of 16S rDNA high-throughput sequencing showed that the microbiota of small and large intestines of Lepus oiostolus comprised mainly Bacteroidetes, Firmicutes, Actinobacteria, Synergistetes and Cyanobacteria, with the genera Enterobacter, Rikenellaceae RC9 gut group, Bacteroides, and Christensenellaceae R-7 group being dominant. A total of 25 bacterial strains were isolated and assigned to 6 genera and 9 species. Drug resistance testing showed that the isolated strains were resistant to 3-27 kinds of antibacterial drugs. Two potential beneficial strains demonstrated certain inhibitory ability against 5 common intestinal pathogens. These preliminary data on the gut microbiome of Lepus oiostolus open the way for further investigation on the microbial ecology as well as microbe-host relationship during health and disease states in Lepus oiostolus.
